DNA Extraction Procedure

DNA or deoxyribonucleic acid is a nucleic acid molecule having a double-stranded, helical structure. This particular molecule is what parents pass on to their offspring; hence it is actually the hereditary material. DNA can be found in any living organism from a unicellular organism to a multicellular one like man. This activity will verify that all living organism have DNA, and that DNA can be extracted from them.

Materials:
Source of DNA (e.g. onion, peas, banana, mango, chicken liver, broccoli) Pineapple juice (fresh) Liquid detergent 95% Ethanol (cold) Salt Water Hot plate Basin Thermometer Ice (2) 250 mL beaker (2) 10 mL graduated cylinder Strainer Blender/Mortar and pestle Stirring rod Knife

Procedure:
1. Prepare a salt-detergent solution by combining the 10 mL detergent, 1.5g salt and water bringing to a final volume of 100 mL. 2. Place the solution in water bath of 50-60oC. 3. Cut the sample into small pieces then add to the salt-detergent solution for. The amount of the sample should be half of the volume solution. Stir gently then let it sit for 5 minutes. 4. Add about 3 mL of pineapple juice in the mixture and keep in the water bath for 10 minutes, immediately placing the beaker of solution in ice bath after the prescribed time. Swirl the solution gently. 5. Macerate the mixture using mortar and pestle. 6. Filter the mixture using the strainer above a 250-mL beaker. 7. Tilt the beaker at a 4o angle then slowly and gently add the ice cold ethanol down the inner wall of the beaker. Allow the mixture to sit for 2-3 minutes. The DNA will move to the top. 8. Place the stick into the first layer of the liquid then gently twirl the stick in one direction only. Lift the stick and the string that will be observed is the DNA.
