Module 3 Protein Trafficking

This document provides instructions for a tutorial on protein trafficking and targeting. It begins with a brief overview stating that proteins synthesized in the cytosol must be sorted and transported to their final destinations, such as organelles, using signal sequences encoded in the proteins. Students are directed to watch a video on early experiments in this area and complete a chart comparing protein targeting mechanisms to different organelles based on lecture information. This will aid in studying for an upcoming quiz on the topic.

Bio/LS 2B03  Tutorial 3  Fall 2010

Protein trafficking and targeting
All proteins are synthesized from mRNAS in the cytosol. Those destined for secretion or for the
various organelles of the cell must be sorted and transported to their final destination. This
involves signals encoded into the proteins that will be recognized by the cellular machinery to
transport the proteins to the correct locations. As was discussed in lecture there are specific
signal sequences that target proteins to particular organelles and specificity can depend on
both amino acid sequence and position within the peptide chain.

For this tutorial you will need to:

1. Watch the video presentation: just the first 6 slides describing the experiments of
Jamieson and Palade, Blobel and Sabatini.

2. Fill in the chart below, use it as a study tool for the quiz and subsequent exams.

3. Complete Quiz 3: Protein targeting, posted on Avenue, due by 8:00am Saturday
October 23rd (worth 4%)

Use this chart to compare protein trafficking/transport to various organelles as you have learned in
lecture.  Note that you should have enough details from lecture to complete the chart for ER,
Mitochondria, and peroxisomes, but you will not be able to fully complete the chart for the organelles
that were glossed over in class.
