Materials:      Mice Diazepam solution Syringe for injection Balance Beaker

Preparation of Diazepam solution: The dose of diazepam solution injected to mice was 25mg/kg body weight. Our ampoule contained 10 mg diazepam in 2mL solution. We took 1mL solution and diluted it with 1mL diluted water. So then each mL of solution contained 2.5mg of diazepam. Let the weight of each mouse is x gm. If the body weight of the mouse is 1000gm, it will be needed 25mg diazepam

=0.025x mg So, 2.5mg of diazepam is present in 1mL solution.

0.025x/2.5mL =0.01x mL solution

So we can see that, if we multiply 0.01 with the body weight, we will get the mL of solution that will have to inject to each mouse. Procedure: 1) 10 mice were taken and they were divided into 2 different groups called test group and controlled group. There were 5 mice in each group.

2) Weight of each mouse of each group was taken and noted. They were numbered by a marker as I, II, III, IV & V controlled group was noted as C1, C2, C3, C4 &C5 and test group was noted as T1, T2, T3, T4, T5. 3) The mice in the control group were injected 0.01mL/gm body weight solution of normal distilled water intraperitoneally. 4) The mice in the test group were injected 0.01mL/mg body weight solution of diazepam solution intraperitoneally. 5) The time of injecting diazepam was noted. The mice were carefully observed. When they got a sleep, we recorded that time from the data. We got latent time or onset of action. We continued our observation and when they awoke-up from the sleep, we also noted that time. From this data we got the sleeping time. 6) The average of latent time and sleeping time were measured. Standard deviation and standard error mean were also determined.

Observation: The mice which were injected with water didnt sleep. So, we can say that diazepam has CNS activity.
