Question: Can we see the difference between Plant and Animal cells under a
If we can, what differences are there?

Background Information: Read this information, Answer the questions


below. -Plant and Animal cells are both Eukaryotic (Eu= true, Karote= kernel) meaning they both have a Nucleus. -Plant cells have a rigid Cell Wall and often have a similar size and shape. Animal cells are more Asymmetrical -Plant cells also have chloroplasts, a organelle with green pigment that converts sunlight to sugar. Finally, Plant cells have a large central vacuole that stores water for the cell.

-Microscope magnification is determined by multiplying the power of the Eye


Piece lens by the Objective lens. Example: An eye piece with 5x power and a objective lens of 10x power.

Questions:
1. What type of Cell are both Plant and Animals?

2. What are 3 characteristics of plant cells that you can see under a microscope?

3. What is the Magnification of an eye piece of 10x and an objective lens of 5x?

Hypothesis- When using a compound light microscope to identify cells, if the

Procedure- Part 1 (indentifying the parts of a microscope)


-Use the labels on the lab station and the diagram below to lable your microscope. (The Instructor or TA must sign-off that you have completed this task with 100% accuracy.)

Procedure Part 2: Making a wet mount slide and operating a Compound


Light Microscope.
1. Cut a letter e out of a newspaper or magazine. 2. Make a wet mount Slide Place the letter e in the middle of the slide. Place a drop of water on top the letter e. Place a cover slip over the letter. 3. Take data. A. Make 3 Circles on a separate sheet of paper with a Petri dish. B. Mark them with the total magnification at the top of the circle. C. Using the microscope, draw what you see at each magnification level. There should be 3 data pictures for each sample.

Procedure Part 3- Animal Cells


1.Gently rub a stir stick on the inside of your cheek or at least 15 seconds. 2.Gently rub it ONE time on the slide. Creating a smear. Throw away the stirrer. 3. Gently place the cover slip gently over your smear. 4.Add a drop of iodine stain to the edge the cover slip. Tap gently on the cover slip with the eraser of a pencil to remove air bubbles. 5.Place the slide on the stage of the microscope. 6.Take Data (see procedure in part 2)

Procedure Part 4- Plant Cells


1. Obtain a prepared slide with onion skin from the teacher. 2. Gently place a cover slip over the onion skin. 3. Add a drop of Metholine Blue to the edge of the cover slip. Gently tap the cover slip with a pencil eraser to remove bubbles. 4. Place the slide on the stage of the microscope.

5. Take Data. (see procedure in part 2)

Conclusion: Write a 3 part conclusion for the question and hypothesis above.
(A 3 part conclusion includes; describing the data in words, your analysis of the data and a statement of the validity of you hypothesis (was your hypothesis correct) and why.

1. Why was it necessary to stain the cells?

2. What is the magnification of: a. an eye piece of 10x and an objective of 4x? b. an eye piece of 10x and an objective of 10x? c. an eye piece of 10x and an objective of 40x? 3. Complete the Venn Diagram comparing Plant cells and animal cells using these terms. (you may have to do some outside research) Many small Vacuoles, Central Large Vacuole, Cell Wall, Cell Membrane, Nucleus, Chloroplast
