Research Work and Interest:


Mechanisms of nerve-injury induced pain:
Peripheral nerve injury in both human and animal models may lead to neuropathic pain. This
type of pain represents a major therapeutic challenge because conventional medical and
surgical pain therapies are ineffective, largely because the pathophysiology is poorly
understood. There is a growing body of evidence which indicates that chronic pain in general
and especially neuropathic pain show a dramatic increase incidence in the elderly population.
In young rats, injury or section of the sciatic nerve promotes sprouting of sympathetic nerve
fibres into dorsal root ganglion (DRG) where they abut the cell bodies of sensory neurons.
Noradrenaline (NA) released from these fibres excites the DRG neurons and facilitate the
generation of spontaneous activity in damaged sensory neurons. This activity is perceived as
pain. The invasion of uninjured DRG by sympathetic axons occurs naturally with age, and the
sympathetic density after peripheral nerve damage is greater in aged than young rats. However,
it is not known how axotomy alters the excitability and sensitivity of aged DRG neurons. It is
of scientific and clinical interest to understand the cellular mechanisms of neuropathic pain in
the elderly. We are testing the hypothesis:
Axotomy-induced changes in physiological and pharmacological characteristics of various
DRG cell types in the young rats will be seen to greater extent in the aged rats.
Using axotomy (sciatic nerve section in rats) as animal model for neuropathic pain, we are
testing this hypothesis by addressing the following questions:
1. Do the characteristics of action potentials and various ion channel conductances of
different DRG cell types change with aging?
2. What are the effects of axotomy on the characteristics of action potentials and various

ion channel conductances of DRG cells in aged rats?


3. Do normal aging and axotomy change the sensitivity of different DRG types to various
neurotransmitters?
Whole-cell patch-clamp recording techniques will be used to address these questions.
Understanding the cellular mechanisms underlying the chronic pain syndromes that are invoked
by nerve injury in the elderly is crucial for designing an effective treatment for these conditions
and for the improvement of their quality of life.
