There are five pages (AA-EE) to this exam, each with two or three
questions per page. For each page of the exam, choose ONE and
only one question to answer. Indicate which question you are
answering by circling its number.
Each question will be scored out of 20 points, so be sure to answer
five questions.
Answer all parts of the questions you choose. Read the question
carefully to make sure you are addressing it, not some other
question you may prefer to answer. Irrelevant information will not
score any bonus points.
You should be able to write your answer on the front of the page,
below the questions; if you absolutely need more space, you may
use the back of the page.
In some cases, you may find it useful to make a small clear drawing to
illustrate and clarify your answer. If you do so, please label the
components of the drawing.
Do not mix up transcription and translation they are both subjects of
this test.
You will have 75 minutes (assuming you arrive on time) for this exam,
so do not spend too long on any single question, at least until you
have answered all of your chosen ones.
If you have time, you may earn up to three bonus points on page FF by
suggesting a change in the content or presentation of this part of
the course (transcription and translation) that would improve it in
the future. (Its only a maximum of 3 points, so dont sacrifice

answering a 20-point question if you cant spare the time you will
get a chance to comment in the course evaluations at the end of
the semester.)

A1. Describe all the elements of an E. coli promoter that may


contribute to regulation of its transcription, and the interactions
the elements have with RNAP.

A2. What is the role of ppGpp in E. coli? When and how is it made, and
for what purpose? How does it affect transcription?

structure) that measured the length of the ssDNA in an open
promoter complex.

C1. The trp operon has two methods of regulation in E. coli. Describe
the mechanisms involved in both methods.

Why is regulation of gene expression at the translational level so
much more rare in prokaryotes?

D2. Explain the differences in the mechanisms of ribosome binding to
mRNA in prokaryotes vs. eukaryotes. Make sure to state any
factors and ribosomal sites that are involved.

D3. Explain how poliovirus takes over translation in human cells, i.e.,
how it slows or stops host translation and promotes transcription
of viral mRNA.

E1. Eukaryotes hydrolyze two GTPs for each amino acid added to a
growing polypeptide. How and why are these two GTPs
hydrolyzed? (Name the factors and identify the specific steps they
promote.)

with respect to each other, and what is the experimental evidence
that this rotation plays a necessary role in translation?
