Lab Write Up

The document summarizes a lab experiment to analyze alu repeats in DNA. The purpose was to see alu repeats and familiarize with lab equipment. Gel electrophoresis was performed but no clear bands were observed, suggesting an error occurred. Possible sources of error and ways to improve the experiment are discussed.

Alu-Repeat Lab

Purpose: This labs purpose was to see the alu repeats in our DNA and helped us get
familiar with the lab and its equipment.

Hypothesis: If we do this lab correctly, Then we will be able to see how many alu
repeats we have.

Procedure: We used the Alu PV92 PCR Lab Procedure by BABEC

Data/Observations:
Below is a picture of our 2% agarose gel with our DNA in it.

One of the things we observed was the lack of results and the presence of fingerprints
on the bottom right corner.

Analysis:
- The graph is labeled vertically with 1-8 and horizontally with A and B. My DNA
was located at point A4.
- There are no lines on the gel that suggests that this lab worked, but there is a
golden light in between line 5 and 6, possibly suggesting that this lab wasnt a
total failure.
- Our results neither proves nor disproves our hypothesis, as we all have DNA, but
this test suggests that we dont.
- One possible factor to error was the use of ice cubes instead of crushed ice,
making the cooling process of our DNA null. The DNA wasnt getting cold
enough because of the ice cube.
- This lab could be improved by using crushed ice instead of ice cubes.
- I am interested in knowing what went wrong.

+/- 10 10 10

-/- 12 0 24

+ Allele Frequency: 54%

- Allele Frequency 46%
+/+: Expected frequency: (.54)^2 = 0.2916.
Actual frequency: 0.4054
+/-: Expected frequency: 2(.54)(.46)=0.4968
Actual frequency: 0.2903
-/-: Expected frequency: (.46)^2=0.2116
Actual frequency: 0.3243

This maths show that we have not reached equilibrium.

Conclusion:
We discovered that there was an error somewhere in our process. The purpose
of our experiment was to find how many alu repeats we had. We did this through
processes such as PCR and electrophoresis. Our evidence is the photo above. As you
may see, the only result we got was the realization that we messed up somehow.
