MATERIALS AND METHODS

3.1 Aquatic Insect Sampling and Measured Water Depth and Canopy Cover

Lata Kinjang and Kuala Woh were selected as sampling sites. These first to second order rivers of

different substrate types and compositions are subject to various anthropogenic disturbances, and

their water surfaces are shaded by a varying amount of canopy cover using tissue roller holder.

The canopy cover of both places was observed. At the sampling site, the river is rather wide with

a relatively flattened river bed made of course sandy substrate and with an open canopy. All

sampling points are located in a running area of fast flowing water. We measured canopy cover

for both two places in three days and the results were recorded. For three days results, we

calculated the percentage of canopy cover by using mean product.

As for water depth results, we used water level tape to measure the depth of water. Also, we record

the results of water depth for both places in three days. The aquatic net was dragged for a distance

of 5 meter underwater and the net was hold upright with on the bottom and open upstream where

the aquatic insects were collected under rocks, dried leaves and twigs. The aquatic insect sample

were placed on the small and large bottle. Small aquatic insects was kept in the small bottle while

the big bottle was used to keep the medium size insect. Some aquatic insect were directly captured

by hand from the river.


The content of each sample from the net was transferred into a properly labelled bottle container.

All aquatic insects were preserved with the small aquatic insects was identified by using

microscope in laboratory. Aquatic insects were counted and kept and properly labeled universal

and polyethylene bottle containing 70% of alcohol or known ethanol. The aquatic insect were

observed under image analyzer and dissecting microscope.
