This chapter presents the results of testing the antibacterial effects of sambong leaf infusion against Staphylococcus aureus. Laboratory tests found the leaf infusion did not inhibit bacterial growth, with a mean zone of inhibition of 7 mm, while the positive control vancomycin strongly inhibited growth, with a mean zone of 23 mm. Statistical analysis using ANOVA found a significant difference between the effects of the leaf infusion and vancomycin. Therefore, the sambong leaf infusion did not show antibacterial properties against S. aureus unlike the positive control vancomycin.

This chapter aims to establish the zone of inhibition of sambong leaf

infusion against clinically isolated Staphylococcus aureus. In doing so,

standard laboratory procedures were done and results were presented in

tabular forms in comprehensible manner. Statistical analysis was

conducted to determine if there were significant difference in the zone of

inhibition of the prepared sambong leaf infusion and the positive control –

vancomycin.

Zone of Inhibition

To determine the resistance of bacterial strains against sambong leaf

infusion against Staphylococcus aureu, paper disk method was conducted in

three trials. Results are as follow.

Table 1 shows that in vitro testing on the antimicrobial bioassay of

sambong leaf infusion and vancomycin against clinically

isolatedStaphylococcusaureus. The sambong leaf infusion showed no


antibacterial activity. Whereas the Positive Control - vancomycin shows

that the clinically isolated bacteria is very susceptible. Results above

suggests that the sambong leaf infusion did not inhibit the growth of the

test organism unlike the positive control. Thus, the plant extract may not

have exhibited a property that will inhibit the bacterial growth. The

exhibited zones of inhibition were interpreted as stated in Kirby-Bauer:

Antibiotic Sensitivity (LibreTexts, 2016).

Statistical Test

To determine if there is an existing significant difference on the

zones of inhibition of the sambong leaf extract and vancomycin against

clinically-isolated Staphylococcus aureus, One-Way ANOVA was utilized

and results is shown in Table 2.

Statistical analysis clearly indicated that there is an existing

significant difference (α<0.05) on the zones of inhibition of the sa,bong leaf

extract against clinically-isolated Staphylococcus aureus when compared

with the positive control. The findings account to the potential of positive

control as the most potent antibacterial agent against clinically-isolated

Staphylococcus aureus as compared to the prepared sambong leaf infusion.
