1. Background
Many reports show that preservatives are added to cosmetics to prevent the
degradation of products by microbial contamination, for example parabens. Some
studies claimed that parabens used by in preservation are deleterious to human health.
Then 1,2-alkanediols have been added in cosmetics because of its low-toxicity and
water solubility. Based on the previous studies, the antibacterial activity of 1,2-
alkanediols against the human skin microbiome such as Staphylococcus aureus and
Staphylococcus epidermidis is effectively used. The writers focused on examining the
effect of the carbon number of the alkyl chain length after the previous revealed that
the molecular structure and the antimicrobial activity of 1,2-alkanediols are useful in
controlling skin microbiome.
In this study, the writers using minimal inhibitory concentration (MIC) and
minimum bactericidal concentration (MBC) to show the effectivity of 1,2-alkanediols
as antimicrobiome.
2. Purpose
1. Evaluating the antibacterial effect of 1,2-alkanediol using MIC with the increase
in the alkyl chain.
2. Evaluating the antibacterial effect of 1,2-alkanediol using MBC with the increase
in the alkyl chain.
3. Methods
The writers using experimental study and describing as quantitative
descriptive. They added the number of alkyl chain of 1,2-alkenediol into 1,2
butanediol; 1,2-pentanediol; 1,2-hexanediol; 1,2-octanediol; 1,2 decanediol; and 1,2-
dodecanediol. The medium containt were used as following components: beef
extract, polypeptone, sodium chloride, and water which had been sterilized in an
autoclave.
The evaluation of MIC and MBC was performed three times for all conditions
to confirm reproducibility. S. aureus and S. epidermidis were cultivated in some kinds
of mediums. To evaluate the MIC, the writers used dilution method to prepare the
resulting sample as mentioned before. Then they took 1000 microlitre of a liquid
medium containing those components and mixed in a test tube. In cultivating phase,
S. aureus and S. epidermidis were shaken by a microplate shaker with different speed
and incubated in different duration among each others. To evaluate the MBC, the
writers comprised 1,2-hexanediol, 1,2-octanediol, 1,2-decanediol, and 1,2-
dodecanediol on agar medium of agar powder and they revealed the most effective
concentration of those substances as bactericidal agents.
4. Results
4.1 Antimicrobial evaluation of 1,2-alkenediol against S. aureus and S. epidermidis
They measured the optical density of the liquid medium including S.aureus and S.
epidermidis which was added to the 1,2-alkanediol solution and incubated for 24
hours (S. aureus) and 48 hours (S. epidermidis). It revealed that the turbidity did
not increase when some 1,2-alkanediols exceeded. The result shows that the MIC
of 1,2-dodecanediol is the most minimally concentration effective and 1-2-
decanediol ; 1,2-octanediol; 1,2-hexanediol in order to the lower turn the higher
concentration. The, the highest concentration needed is raised by 1,2-butanediol
and 1,2-pentanediol which are revealed that they are not effective in antimicrobial
activity.
4.2 Bactericidal evaluation of 1,2-alkanediol against S. aureus and S. epidermidis
MBC was measured for 1,2-hexanediol, 1,2-octanediol, 1,2-decanediol. The result
shows that the MBC of 1,2-decanediol exhibited bactericidal activity at the most
minimally concentration needed then in order 1,2-octanediol to the second most
minimally concentration. In contrast, 1,2-hexanediol and 1,2-dodercanediol did
not exhibit significant bactericidal activity.
5. Conclusion
The conclusion of this experimental study is revealing that the addition of the
alkyl chain in 1,2-alkenediols can increase the effectivity both antimicrobial and
bactericidal activity. But, there were any obstacles stated in methods part of the
journal that it is desirable to perform centrifugation prior to the evaluation of MBC
but it was not performed because there were not enough samples.
6. Superiority
The writers explained the mechanism in detail in resulting the appropriate
result in purpose.
7. Shortcoming
Based on the obstacles, there were not enough samples that made the writers
could not perform centrifugation prior to the evaluation of MBC even the result was
not significantly affected.
