Alternate Photosynthesis Lab assignment and Lab Quiz information

Dear student

I’ve created an alternative online assignment for you as I do not have the resources at home to
reproduce the photosynthesis lab. I selected a few video demonstrations that can be used to
replicate some portions of our photosynthesis lab. You can find all these videos on Canvas in
the Lab Module or you can click the links in this document to view the videos.

Instructions:
1) Please view then answer the follow-up questions to the video presentations.

2) Type in your answers on this document and submit them on your designated lab date.

These questions will also prepare you for the lab quiz, which you will take on your designated
lab day as well.

1) Which gas is required by the Calvin cycle? which gas is produced by the light reactions?

The gas required for the Calvin cycle carbon dioxide, the gas produced by the Light reaction is oxygen.

2) The reaction above shows the reactants and conditions required for photosynthesis as well
as its products. Which leaf disk treatment group contained all that is necessary for
photosynthesis?

The leaf group that contained all components necessary for photosynthesis to occur was Group #1 which was under the light.

3) What is the source of CO2 in this experiment?

The source of CO2 in this experiment is the sodium bicarbonate that was added to the beaker of water.

4) Why do leaf disks float when they receive light, but not when placed in the dark? Your
answer should include specific reactions of photosynthesis.

Leaf disks float when they receive light because the light commences the Light Reactions which produce Oxygen making
them float. The leaf disks do not float when placed in the dark because they are not exposed to light, not allowing the light
Reactions to commence.

5) What do you predict would happen if both leaf disk groups received light, but one received
water and the other received sodium bicarbonate?

I predict the leaf disk group with water would float because H2O and Light are required to commence Light Reactions.

photosynthetic pigments using Paper Chromatography.

Follow up Questions:

1) In your own words, describe paper chromatography? (you may also read about it on pg 65
of your manual.

Chromatography is the separation of pigments from a mixture. A stationary phase holding drops of the mixture is partially submerged
into a mobile phase which is a fluid that has the parts of the mixture separate showing all parts on the stationary phase as they separate.

2) What is the source of the photosynthetic pigments in this video. Specifically what part of
the chloroplast are they from?

The source of the photosynthetic pigments are from the chlorophyll within the thylakoids in the chloroplast.

3) Stop the video at minute 5:20, then go to step 20 at the bottom of pg 68 of your lab
manual. Read the description of the photosynthetic pigments and their relative position on
the paper chromatogram at minute 5:20. List below the order in which these pigments (in
step 20) appear starting rom the bottom of the paper.

1.Chlorophyll b
2.Chlorophyll a
3.Xanthophyll
4.Carotene

4) Which pigment is missing from the paper?

The missing pigment is anthocyanin.

5) What vegetable could we have added to the green leaves to get the missing pigment?

A vegetable that could have been added was cauliflower or a potato.

Follow up Questions:
It is important to understand that pigments can absorb and transmit different wavelengths of
visible light.

1) What color(s) are absorbed by the green filter?


All colors besides green are absorbed by the green filter as green light is
transmitted through.
2) What color(s) are transmitted by the blue filter?
All colors besides the blue are absorbed by the blue filter as blue light is transmitted through.
3) Summarize the effect the colored filters had on the transmission of light.
Colored filters absorb distinct colors from the transmission of light to emphasize one color. A red
color filter will absorb all colors besides red to transmit red light through the filter.
