Activity No. 1 Microscopy
2nd Quarter
Introduction
Scientists use different tools to study about life. These tools help them perform experiments efficiently
and effectively. One of the most useful tool in studying science is a microscope. A microscope is an
instrument used to examine minute objects, structures and organisms that are too small to be seen with the
naked eye. The microscope has three sets of functions: mechanical, illuminating and magnifying.
Using the compound microscope seems like a simple laboratory task. In this activity, you will learn
techniques and important guidelines that student must be familiar with in order to fully optimize its use.
These techniques are also important in protecting one’s eye from strain when examining objects under the
microscope for a long period of time.
Objectives
 Identify the parts of the compound microscope and explain the function of each part
 Use the compound microscope properly
 Magnify a specimen using the low-power and high-pawer objectives
 Compute for that total magnification of an object under the microscope
Materials
Cover slips Hair strand
Glass slides Small letter “e” (cut out from a new paper)
Compound microscope Red thread
Scissors/cutter

Procedure

A. Using the Compound Microscope


1. Obtain the compound microscope. Hold it with both hands, one in its arm and the other under the
base
2. Place the microscope away from the edge of the laboratory table, do not drag the microscope, lift it
up if you need to adjust its position
3. Turn the revolving nosepiece and align the low power objective with the body tube
4. Look at the microscope from the side. Lower the body tube by turning the coarse adjustment knob
until the objective lens tip is about ½ in the surface of the stage
5. Look into the eyepiece. Adjust the mirror so that the field being observed is well illuminated. Tilt the
mirror until you are capable to capture uniform light
6. Adjust the diaphragm opening to regulate the intensity of light
B. Procedure in Observing Specimen
1. Mount the specimen (hair strand, thread, cut out of letter “e”, onion) on the slide and cover it with
cover slip
2. Place the slide with the specimen at the center of the stage of the microscope. Adjust the slide with
the specimen at the center
3. While looking into the eyepiece, slowly turn the coarse adjustment knob to raise the body tube until
the object comes into view. Continue adjusting the position of the body tube until the specimen is
clear in focus. This vie wis in LPO. Make a sketch of your observation
4. Shift the magnification to high power objective. Readjust the view carefully using the fine
adjustment knob. Sketch the HPO view of the your observation.
