Effect of pH and Temperature on Enzymatic Activity

Introduction:

Biological catalysts called enzymes increase the rate of reactions without


themselves being chemically changed. Enzymes are globular proteins folded into a
complex 3-dimensional shape that contain a special surface region called the active site
where a specific substrate binds structurally and chemically. Enzymes speed up
reactions as they lower the activation energy, which is the minimum energy barrier
needed to be overcome before a reaction can occur by providing an alternative reaction
pathway.

In this experiment, the enzyme that will be used is catalase. This molecule is
commonly found in animal and plant cells, but a substantial amount is found in potato.

The substrate that will be catalyzed is hydrogen peroxide (H2O2) which is a poisonous
waste product in the human body. Therefore, the catalase enzyme will specifically
catalyze the hydrolysis reaction of H2O2 into safe, non-poisonous products of water and
oxygen gas:

2H2O2 ⟶ 2H2O(l) + O2(g)


Problem:

How could certain factors such as pH and Temperature affect enzymes?

Hypothesis:

pH: pH could affect the activity of the enzyme (catalase),this enzyme works best at pH
between 6 and 8.If the environment is too acidic or too basic compared, the enzyme will
denature and will not be able to work properly.

Temperature: Temperature could affect the activity of the enzyme (catalase). The
reason is because if the environment is too hot or too cold the enzyme will have trouble
getting accustomed to either temperature. After it will then get denatured and stop
functioning.

Materials:

Potato juice, test tubes, test tube rack, hydrogen peroxide, metric ruler, ice bath, boiling
water bath, pipettes,distilled water, NaOH, HCL, water, 6 test tube, holder, 10 mL
graduated cylinder, ph meter, goggles, and gloves.

Procedure:

Part A: pH

1. Put 5 drops of potato juice in the three test tubes labeled A.


2. Label each test tube A1, A2, and A3.
3. In the first test tube, add 5 drops of distilled water to create a neutral medium.
4. In the second test tube, add 5 drops of NaOH to create a basic medium.
5. In the last test tube, add 5 drops of HCL to create an acidic medium.
6. Add 3 mL of hydrogen peroxide(substrate) to each test tube.
7. Wait 2 minutes and measure the foam layer using a ruler for each test tube.
8. Record the results in the table.

Part B: Temperature

1. Put potato juice in 3 test tubes labeled B.


2. Label each test tube B1, B2, and B3.
3. Leave the first test tube in the test tube rack(room temperature).
4. Put the second test tube in the ice bath(approximately 0 degrees).
5. Put the last test tube in the hot water(approximately 100 degrees).
6. Wait 2 minutes then take them out.
7. Add 3 mL of hydrogen peroxide(substrate) to each test tube.
8. Wait 2 minutes and measure the foam layer using a ruler for each test tube.
9. Record your data in the table.
