4. Connect the power cord. If the grounding is not very good (grounding wire and
zero wire voltage is higher than 0.5V DC), please connect the grounding cable
to the grounding pole and connect the other end to the ground of the lab.
5. Then turn on the analyzer.
Part II Programming
1. Item: Go to Main MenuProgramItem. Click Set to edit item.
2. Method: Mostly we use three kinds of test method, End-point, Fixed time and
Kinetics. Please select according to the reagent specification.
End-point: After all reagent and sample are mixed together which is incubated
outside of the machine, the instrument reads only one absorbance value to
calculate the sample concentration.
C sample = A sample /A calibrator × C calibrator
A: Absorbance value C: Concentration
Fixed time: After all reagent and sample are mixed together, the machine
reads two absorbance values to calculate the sample concentration.
Kinetics: After all reagent and sample are mixed together, the instrument read
at least three absorbance values to calculate the sample concentration.
C sample=ΔA sample /min /ΔA calibrator /min × C calibrator
ΔA/min: absorbance changing ratio C: Concentration
3. WL: WL means how many wavelengths used for this item. Refer to the reagent
specification to set how many wavelengths are needed.
4. TEMP: TEMP means temperature. Refer to the reagent specification to the
reaction temperature. There is 25℃, 30℃, 37℃ and RT (room temperature)
for selection. We suggest to use 25℃ instead of RT for testing.
5. ASP VOL: Set it as reagent instruction requested. It is suggested to set at least
500ul. If total volume of reagent and sample is less than 500ul, please increase
reagent and sample volume by percentage.
6. DEC and Unit: DEC means decimal and Unit means test result unit. Set it
according to reagent specification.
7. Blank: There is Reagent blank, Sample blank and No for selection.
8. WL: WL means filter wavelength. In the above setting, there is also a WL
setting. If you set WL as 1, please choose one filter, for example 340nm. If you
set WL as 2, please choose 2 filters, for example 340nm and 630nm.
9. Points: Points mean how many absorbance values need to read. It is
automatically set when you select Method.
10. Normal, General, Male and Female: Set reference range according to reagent
specification.
12. Calibrate: Select the calibrate method. For reagent with one calibrator, please
select Linear-1P. For reagent with more than one calibrator, please select
accordingly. If reagent kit does not include calibrator and the test method is
Kinetic, please select No STD.
13. Linear Range: Set it according to reagent specification.
14. Delay Time: Delay time helps to ignore the instable absorbance when reagent
and sample start to reaction. For End-point method, reagent specification may
not indicate the exact time and we suggest set it as 5-10sec. For Fixed time
and Kinetics, please follow the reagent specification to do setting.
15. Interval Time: Interval time is only available for Fixed time and Kinetics. It is
the reading interval between the two absorbance reading.
16. STD Num: STD Num means the number of Calibrator (Standard).
17. STD: Input the concentration of the Calibrator. For multi-calibrator, please input
STD1 and STD2.
18. Factor: The Factor is used to calculate sample result. If you don’t edit it, after
reading the absorbance of Calibrator, it will automatically calculate the Factor. If
you input it manually, it is not necessary to test Calibrator. For Kinetics, most of
the reagent specification will indicate the Factor and you can input directly.

This is a part of reagent instruction. We can know it needs only one absorbance value
after mix all reagents and sample. From the calculation formula, we can find that it is the
End-point test method. For blank, it needs R and Saline water (no sample is needed), so it
is used Reagent blank. Please find the detailed programming as below.

Dissolve one bottle of R1 with a ration of R2 (refer to label onR1) as working reagent.
Allow the working reagent to equilibrateto the reaction temperature prior to processing.
Calculation:
ALT (U/L) = (ΔA Sample/min–ΔA blank/min) × K
K = Vtotal / (Vsample×extinction coefficient) = 1746
Expected Value: Male ≤40 U/L Female ≤35 U/L

Mix and incubate at reaction temperature for 60 seconds. Read the initial absorbance and
the change of absorbance in every minute for 3 minutes separately. Determine the change
of absorbance per minute (ΔA/min). ALT (U/L) = (ΔA Sample/min–ΔA blank/min) × K.
This is a part of the reagent instruction. We can know, it needs 3 absorbance values. From
the calculation formula, we can find that it is the Kinetics test method. The detailed
programming is as below.

2. Wait for the preheating time. Generally, it needs 1-2minutes to wait until the
lamp and heating are stable.
3. Click Measure to start the test. Then follow the message on screen to aspirate
Rinse, DI water, Blank, Standard and Sample. Then the analyzer will show and
print the result.
