METHYLENE BLUE REDUCTION TEST (MBRT)

Aim: To check the quality of different milk samples.

Principle:
Large number of bacteria present in raw milk especially coliforms are indicative of poor methods
of production and handling. Bacteria like Escherichia coli and Streptococcus spp. utilize the
oxygen present in milk thereby producing a reduced environment. This reduced environment can
be detected by Oxygen Reduction Test.
In this test, 1% Methylene Blue solution is treated as an indicator solution. Under the presence of
oxygen, Methylene Blue shows its characteristic colouration. Under reduced condition, Methylene
blue accepts hydrogen and itself gets reduced to leuco or colourless form.
The rate of oxidation-reduction depends on the rate of growth of microbes at 37°C in the milk with
methylene blue solution.

Materials Required:
• Milk Samples i.e.–
1. Sample I: Pasteurized milk opened in sterile conditions under laminar hood.
2. Sample II: Boiled milk
3. Sample III: Raw milk brought from a cow shed.
4. Sample IV: Milk was boiled and then cooled and kept at room temperature for 4-5
hours.
• Methylene Blue solution.
• Culture tubes, cotton plugs, Pipette, water bath.
Procedure:
1. To 9 ml of milk sample, 1 ml of 1% Methylene Blue solution is added under sterile
conditions.
2. The tubes are shaken properly and incubated in a water bath at 37°C.
3. After every 2 hours, the colour of the incubated milk samples were observed. Any change
in colouration from blue to leuco in any of the samples was noted.

Observation:
