Dimethyl sulfide (DMS) is a polar, aprotic organic solvent that is commonly used as a cryoprotectant to prevent freezing damage to tissues. DMS reduces ice formation and cell death during freezing by preventing crystallization. Cell viability assays, such as the resazurin assay and MTT assay, are commonly used to investigate cell response to drugs and determine if cells are alive or dead. The MTT assay depends on the ability of NADH-dependent oxidoreductases in living cells to reduce the tetrazolium salt MTT to form an insoluble purple formazan product. This allows quantification of viable cells based on the amount of formazan produced.
