1 SCOPE AND FIELD OF APPLICATION
This International Standard specifies a reference method
for the determination of the nitrite content of meat and
mest produots.

3. DEFINITION

4 PRINCIPLE

5 REAGENTS
All reagents shall be of analytical quality. The water used
shall be distilled water or water of at least equivalent
purity.
5.1. Solutions for precipitation of proteins
5.1.1 Reagent 1
Dissolve 1069 of potassium ferrocyanide trihydrate
[KaFe(CN)g-3H20} in water and dilute to 1 000 mi
6.1.2 Reagent It
Disolve 2209 of zine acetate dihydrate
[Zn(CHsCOO),-2H,0] and 30 mi of glacial acetic seid in
water and difute to 1 000 mi
61.3 Borax solution, saturated
Dissolve 504 of disodium tetraborate decehydrate
(N22B407-10H20) in 1 000 mi of tepid water and ooo! ta

5.2. Sodium nitrite standard solutions
Dissolve 1,000 g of sodium nitrite (NaNO2) in water and
gilute to 100 mI in one-mark volumetric flask. Pipette
SmI of the solution into a 1 000 ml one-mark volumetric
flask, Dilute to the mark.
Prepare a series of standard solutions by pipetting 5 mi,
10ml and 20 mi of this solution into 100'mi one-mark
volumetric flasks and diluting to the mark with water
These standard solutions contain respectively 28 ug, 5010p
and 10,0 ug of sodium nitrite per miliitre,
‘The ‘standard solutions and the dilute (0,05 g/l) sodium
nitrite solution from which they are prepared shall be made
up on the day of use,
5.3 Solutions necessary for colour development
8.3.1 Solution |
Dissolve, by heating on a water bath, 2g of sulphanilamide
(NH2CsH«SOgNH2) in 800m! of water. Cool, filter, if
necessary, and add 100 mi of concentrated hydrochlorie
acid solution (p29 1,19 g/mil, while stirring. Ditute to
1.000 mi with water.

Dissolve 0,25 9 of (W-1-naphthylethylenediamine dinydro-
Shloride (CyHy NHCHCHNHa-2HCI) in water. Dilute to
250 mi with water
Store the solution in a wel-stoppered brown bottle. It shall
be kept in a refrigerator, for not longer than one week
