INTRODUCTION
Lipids are biological molecules that are insoluble in water but soluble in nonpolar solvents. It
constitute a very important heterogenous group of organic substances in plant and animal tissues, and
related either actually or potentially to the fatty acids.

LEARNING OBJECTIVES:
At the end of the experiment, the students must be able to:
1. Identify the physical and chemical properties of fats and oils.
2. Detect kinds of lipids using Qualitative tests.

TESTS:

II. SOLUBILITY TEST
III. TEST FOR UNSATURATED GLYCRIDES

MATERIALS:
1. Test tubes 5. Spatula 9. Vegetable oil
2. Test tube rack 6. Filter paper 10. Coconut oil
3. Droppers 7. Distilled water
4. Filter paper 8. Margarine

REAGENTS:
1. Acetic acid 4. NaOH
2. Ethanol 5. HCL
3. Hexane 6. Alcoholic Iodine

I. PRACTICAL TESTS FOR FATS AND OILS


• TEST SAMPLES:
a) Acetic acid c) Vegetable Oil
b) Margarine d) Ethyl Alcohol

PROCEDURE
1. Cut a filter paper into 4 and label them with the samples to be tested: Acetic acid, Margarine,
Vegetable Oil and Ethyl Alcohol.
2. Prepare droppers and spatula. Slowly place drops of each of the samples and let the filter paper
stand for at least 3 minutes and note which sample left a translucent spot. Find a good source
of light for better viewing of results.

TEST WITH DESCRIPTION OF RESULTS INFERENCE


Acetic acid

Vegetable Oil

Ethyl Alcohol

✓ Expected result: presence and absence of translucent spots.

II. SOLUBILITY TEST
• TEST SAMPLES:
a) Hexane d) Water
b) Ethyl alcohol e) HCL
c) NaOH

PROCEDURE
1. Prepare 5 test tubes. Place 1mL of the following solvents in a separate test tube: hexane, water,
ethyl alcohol, HCL and NaOH. Make 2 sets each.
2. In the first set of the solvent, add drops of coconut oil. Shake well and observe solubility.

✓ Expected result: Miscible and immiscible

3. In another set of solvents, add a pea sized amount of margarine each of the 5 test tubes. Shake
well and observe solubility again.

III. TEST FOR UNSATURATED GLYCERIDES


• TEST SAMPLES:
a) Coconut Oil b) Vegetable Oil c) Alcoholic Iodine

PROCEDURE
1. Place 10 drops of the coconut oil to alcoholic iodine in a test tube. Shake well and observe for
color change.
