Cell Respiration Lab

This document describes a laboratory experiment to measure the rate of cell respiration in germinating seeds at different temperatures. Seeds are placed at 40°C, -4°C, and room temperature in a respiration chamber with a CO2 sensor, and CO2 production is measured over 5 minutes at each temperature. The experiment aims to study the effect of temperature on respiration rate and provide evidence that respiration occurred in the seeds by producing CO2.

01 Cell Respiration

Laboratory activity

Cell respiration refers to the process of converting the chemical energy of organic molecules into a form
immediately usable by organisms. Glucose may be oxidized completely if sufficient oxygen is available
according to the following equation:

C6H12O6 + 6O2(g) → 6 H2O + 6 CO2(g) + energy

All organisms, including plants and animals, oxidize glucose for energy. Often, this energy is used to
convert ADP and phosphate into ATP. Seeds undergo cell respiration during germination.

OBJECTIVES

In this experiment, you will

• Measure CO2 production.

• Study the effect of temperature on cell respiration.

• Compare the rates of cell respiration od seeds in different temperatures.

MATERIALS

- Computer or phone with Vernier Graphical analysis app.

- Germinating seeds (prepare 3 equal wight containers)
- Respiration chamber
- Vernier CO2 sensor
- Thermostat
- Freezer

PROCEDURE

Using the CO2 Gas Sensor, you will monitor the carbon dioxide produced by seeds during cell
respiration.

1. Prepare 3 equal wight petri dishes with germinating seeds.

2. Place one of the dish in thermostat (40 °C) and another in Freezer (-4 °C). third dish will be kept
in room temperature. Wait 20 minutes.
3. Place the germinating peas into the respiration chamber
4. Place the CO2 Gas Sensor into the respiration chamber shown in Figure 1

Figure 1. CO2 Gas Sensor in respiration chamber

5. Wait one minute for readings to stabilize, then begin collecting data by clicking „Collect“ .
Collect data for 5 minutes and click „Stop“.
6. Repeat this process with all three samples. After each sample clean respiration chamber.
7. Answer questions and prepare laboratory report.

Laboratory report should include:

- Variables
- Hypothesis
- Research question
- Graphically presented results
- Conclusion

QUESTIONS

1. Do you have evidence that cell respiration occurred in seeds? Explain.

3. What is the effect of temperature on the rate of cell respiration in seeds?

4. Why do germinating peas undergo cell respiration?

EXTENSIONS. For your IA it is possible to analyse how different variebles affect seed respiration. You
could: compare the respiration rate among various types of seeds; compare the respiration rate among
seeds that have germinated for different time periods, such as 1, 3, and 5 days ect.
