WORKING
1. Switch on the instrument and let the sodium lamp
illuminate to its maximum brightness.
2. Set the alignment of the sodium lamp compartment to the optical bench, D. Ensure
that all the settings are done in the zero-zero position.
3. Open the polarimeter tube compartment and place the polarimeter tube, which is
filled with sample in such a way as to avoid air bubbles
4. Specific attention should be paid to temperature control of the solution and of the
polarimeter.
5. Observations are done by observing through telescope when the half dark and half-
light portions of a circle are in equilibrium.
6. It’s half dark and half-light portion is matched with a knob by rotating it in clockwise
or 2 anti-clockwise directions. It is also defined as dextro (+) or levo (-) rotatory
respectively.
7. Note the observation readings and also perform a blank determination in the similar
manner.

WORKING
1. Open the prism lock and place few
drops of liquid testing sample at the temperature specified
2. Allow the liquid to smear on the lower prism surface quickly close the prism
lock. Adjust the eye piece cross wire with a knob by rotating it in a clockwise or
anti-clockwise.
3. Clockwise direction so that the sharp demarcation line passes through its centre.
4. Sharp demarcation is produced in the form of a circle having half-light & half
dark Portion.
5. Fine adjustment can be done by a knob provided just above the prism.
6. Note down the reading from the scale with the help of the magnified lens of the
Instrument.
MAINTENANCE & PRECAUTIONS
1. Ensure that the instrument is free from dust.
2. Ensure that all the parts including eyepiece., prisms and mirror are clean before
operation
3. Place the instrument on a table where adequate light can be reflected through the
mirror
4. Cover the instrument after use.

WORKING
1. Connect the main cord to the supply
and switch on the instrument
2. Hot plate starts heating when switched on. The heating temperature can be
controlled (normally at 37°C) with temperature control knob
3. Place a water filled glass beaker on the hot plate and wait for water to attain adjusted
temperature
4. Fit tablet basket to the motor shaft and put the tablets into the tubes
5. Put stoppers into the tubes to restrict the tablets within the tubes
6. Press the push button to start up & down motion of the shaft carrying the tablet basket
(Basket starts oscillating in and out of the beaker)
AIM: To evaluate the ability of the tablets to withstand abrasion in packing, handling and
transporting.

WORKING
1. Unscrew the knob from the axle & remove the cover
of tumbling chamber
2. Weigh the tablets to be tested and load them into the
chamber
3. Place the cover with the round knob to lock the
tumbling chamber to the axle
4. Select the time period of rotation from the timer
(normally for four minutes or 100 revolutions)
5. Connect the main cord to the supply and switch on the instrument
6. The chamber starts rotating and stops after the time selected at the timer
(during each revolution the tablets fall from a distance of six inches to undergo shock)
7. When rotation stops, remove the cover and take out the tablets
8. Weigh the tablets and measure the loss in weight that indicates “friability”

WORKING
1. Unscrew the container holder and remove the cap of
cylindrical container
2. Fill the cylindrical container with the material to be ground plus the grinding medium
3. Place the cap and screw back
4. Connect the main cord to the supply and switch on the instrument
5. The cylindrical capped container starts rotating on their axis
6. Adjust the speed of rotation using speed regulator
7. An internal cascading effect between balls is produced that reduces the material to a fine
powder

1. The speed of rotation should be kept equal to or below critical speed (speed after which the
steel balls start rotating along the direction of the cylindrical device causing no grinding)

WORKING
1. Fill empty capsule into the loading tray and place it
over the bed
2. Lock the body of capsules using handle and remove the
separated caps in the loading tray itself using lever
3. Accurately weigh the amount of drug to be filled in the capsules and place it in powder tray
kept over the bed.
4. Spread the powder using powder spreader to fill the bodies of the capsules uniformly.
5. Collect excess of the powder on the platform of the powder tray
6. Lower the pin plate to press the powder in the bodies
7. Remove the powder tray and place the caps holding tray in position
8. Press the caps using plate fitted with rubber top
9. Use lever to unlock the filled capsules in the tray

1. A spherical coating pan (made up of stainless steel) or a
polishing pan (consists of canvas drum) attached to a
motor.
2. A motor housed in a heavy metal enclosure to avoid
vibration during operation.
3. A front panel (bearing on/off button and other controls of
the instrument).
4. Air blowers fitted to pans to force hot or cold air to
facilitate drying.
WORKING
1. Place the tablets to be coated in the coating pan “OR”
tablets to be polished in the polishing pan
2. Connect the main cord to the supply and switch
on the instrument
3. Spray the coating solution over the tablet bed while the coating pan is kept rotating
4. After completion of spraying, allow the tablet bed to dry using hot air blower
5. Remove the tablets from the chamber after proper drying

WORKING
1. Fix clamp B in the back side of tablet machine body A with the bolt tight with the spanner.
2. Put the stainless-steel hopper C in clamp B
3. Fix one part of tablet powder pipe G in the hopper and other part in the tablet machine’s
shoe D
4. Put the heavy wheel E, at right side of tablet machine and fix it with the help of clamp
provided
5. Take the desired powder and fill in the hopper and rotate the wheel in clockwise
direction
6. The tablet will come out automatically at the bottom of Tablet machine’s shoe

WORKING
1. Switch on the mains through stabilizer.
2. Set the regulator knob so as to attain a temperature of 8°C.
3. Ensure that indicators are all functioning properly.
4. Record of the day to day monitoring of the temperature is maintained separately in
a log sheet.
5. Temperature should be between 2° to 8°C.
6. Defrost and clean once in fifteen days.
7. While defrosting the stored materials shifted to another refrigerator

AIM: To heat the sample with a temperature up to 100°C through boiling water.

WORKING
1. Ensure that sufficient amount of water is available in the water bath.
2. Ensure that the instrument is properly connected to power supply.
3. Fill the bath to the level little above the falls bottom
4. Switch on the supply. Put the toggle switch On.
5. Set the thermostat knob to the required temperature and rising in the temperature is
indicated by glowing the indicator lamp.
6. When required temperature is reached, the indicator lamp will go OFF.
7. The bath is then allowed to work.
8. After completion of heating process, reduce the temperature to zero, by rotating the
temperature setting knob to zero position. Switch off the power supply and allow the
bath to cool to the desired temperature before the control knob is turned backwards.
This is to eliminate unnecessary strain on the thermostat.
MAINTENANCE & CLEANING
1. Ensure that the power supply to the instrument is switched “off” before cleaning.
2. Clean the instrument with a clean dry cloth every day.
3. Once in a week, remove adhered dust by wet mopping using detergent solution and
afterwards wipe the surface with a clean dry cloth to remove traces of detergent.
4. Precautions have to be taken to clean the instrument immediately with to remove the
moisture.
5. Change the water in the water-bath once in 2 days and use always Demineralized
water.

WORKING
1. Ensure that the instrument is connected to the power supply.
2. Switch “ON” the main power supply, glowing of red light at mains indicates the power
supply.
3. Switch on the instrument by ON position which leads to activation of green control bulb
and the temperature controller.
4. Set the required temperature by temperature regulator nob
5. The temperature display shows the actual temperature of furnace and the red light of the
temperature controller glows.
6. When temperature reaches at the setting point, the red light of temperature controller
automatically switched “OFF” and green light will glow. The equipment is now ready
for operation.
7. If any discrepancy observed during operation, inform to maintenance for rectification.
MAINTENANCE & CLEANING

CALIBRATION OF VISCOMETER
PROCEDURE
1. Before adding material in the cylinders ensure that the outer and inner surface of the
cylinders are clean arid dry.

measured volume of liquid through orifice of a

WORKING
1. Place the dip tube in the solution to be filled.
2. Direct the nozzle into the sample.
3. Connect the pump to the electrical supply.
4. Switch on the pump.
5. Adjust the quantity to be filled in the ampoule using the dose adjustment knob.
6. Continue the operation till the fills are complete.
7. Switch off the pump.
8. Open the LPG and the oxygen supply knob.
9. Light-UP and adjust the flame intensity by regulating the flow with the help of valves.
10. Place the ampoule in the ampule holder of apparatus.
11. ROTATE the ampoule slowly and ensure proper sealing.
12. Remove the ampule.
13. Switch off .the gas and oxygen supply.

1. A barrel containing a compressible spring held between
two plungers.
WORKING
1. Ensure that the instrument is clean and free from any residue of dust or tablet particle.
2. The lower plunger is placed in contact with the tablet, and zero reading is taken. The
tablet must be placed in inclined cantilevered position.
3. The upper plunger is then forced against a spring by turning a threaded bolt until the
tablet fractures.
4. As the spring is compressed, pointer rides along a gauge in the barrel to indicate the
force.
5. The force of fracture is recorded and the zero force reading is deducted from it.
6. The hardness is taken in kg per sq. cm.
MAINTENANCE & PRECAUTIONS
1. Never keep the spring under tension, when not in use
2. If the instrument does not produce required results or its response is poor then it should
be labelled faulty and should be repaired or serviced.
AIM: To sterilize anything that can withstand a temperature of 121oC for 30 minutes.

WORKING
1. Ensure that there is sufficient water inside the
chamber.
2. Load the autoclave and fasten the lid keeping the discharge tap open.
3. Adjust the safety valve to the required temperature and turn the heat on.
4. Set the desired pressure by adjusting the knob on top of the pressure control switch.
5. When the pressure reached the fixed, the bell starts ringing. The element will gets off
disconnected, when the pressure falls down up to 0.5/ the element will starts
automatically
6. After that you can fix the timer and switch off the mains.
7. After the sterilization release the steam from the steam release system.
8. Slowly open the air and steam discharge taps after the pressure gauge has reached
zero.
9. Allow the material to cool before these are handled (usually agar bottles take hours
before these become safe to handle).

WORKING
1. Ensure that the equipment is clean and power
is supplied through suitable voltage stabilizer
of 1.5KV.
2. Connect the equipment to the main power supply
3. Switch ON the unit.
4. Observe the digital display indicator
5. To set the desired temperature press the “set” touch key and simultaneously the
potentiometer knob turn to right hand side or left-hand side with the help of screw driver
(clock wise to increase the set value or anticlockwise to decrease the set value) release
the ‘set’ touch key
6. If you press the ‘set’ touch key it will indicates the set temperature. If you release ‘set’
touch key it will indicate the current temperature.
7. If the current temperature in the chamber raises above the set value, heating
automatically ‘puts off’ and cooling will be “ON” and the temperature in the chamber
decreases below the set temperature cooling automatically “puts off “and heating will be
“ON”. This process repeats.
8. Ensure that the observed temperature after stabilizing it should not differ ± 2 °C to the
set temperature in the log sheet.
9. Maintain the record day-to-day monitoring of the temperature separately in a log sheet
10. Record the temperature using maximum and minimum thermometer once in a day in a
log sheet.

WORKING
1. Ensure that the instrument is clean and kept in proper position.

3. Switch on the instrument.
4. Place the Petri dish containing bacterial colonies on the wolffhuegel grid glass plate.
5. The instrument is provided with 100 mm diameter magnifying lens and auto pen
marker which is fixed to 3 pin sockets provided on right of instrument.
6. Take the auto marker pen and mark each colony by pressing the pen tip and the Digital
counter will start counting sequentially.
7. The counter can be set to zero by pressing the center push button switch.
8. Counting can also be done manually without using pen marker by pressing the push
button switch provided right hand side of the lower front of the panel.
9. The instrument displays four-digit counters with beep sound for each counting.

MAINTENANCE & PRECAUTIONS
1. The operator should scrub his/her hands thoroughly with alcohol and wear sterilized
gloves, headgear, mask and clothing.
2. All the apparatus and equipment required to be used in the process must be sterilized
in advance.
3. Before operation UV lamp should be on and after the operation the lamp should be off.
WORKING
1. Adjust the desired pressure.
2. Now connect the filtration assembly (Vacuum flask) to the pump.
3. Pour the sample want to filter on the Buckner funnel place above the vacuum flask.
4. Now start the pump by holding the flask.
5. After completion filtration or drying switch off the pump and take out the sample.
WORKING
1. Ensure that balance is aligned properly and cleaned the spirit level should be in the
centre of the circle.
2. Ensure that the display shows 0.00000.
3. Place 4 standard weights of different denominations as per schedule & record the
weight.
4. The observed weight should confirm with the acceptance criteria as given in the
balance log sheet.
5. If the instrument goes out of calibration contact the service person for necessary
adjustment and recalibrate.
The equipment will baseline and second pop up will come up that will instruct you to
block the sample beam, do so by placing the cardboard to block the beam. Once this is
done click OK.
Once the Baseline procedure is done remove the cardboard from the sample chamber.
Click on Zero on the right-hand panel.
Once zero is finished take out the blank sample and put in your sample.
Go to Start at the top center of the screen and click on it. Save your data to the correct
file folder. Click Ok. The scan will begin at this time
Once the scan is finished, a pop up will come up that will allow you to scan another
sample of finish. If you are running multiple samples change your sample, change the
sample name on the pop-up box, and click OK otherwise click Finish.

1. Put the drying agent (calcium chloride or silica gel etc.) in the
lower portion of chamber.
2. Take the substance, put in the chamber and close the box tightly.
3. Wait for some period of time and then taken out the substance

2. Before placing substance check the absorption of drying agent.

6. After the required time take out the beaker, remove the bead from solution.
7. Put off the main switch.

2. Ensure that all the parts including optical lens, telescope lens and polarimeter tube are
clean.
3. Polarimeter tube should be filled so as to avoid any air bubbles.

1. The herbs are chopped or powdered, put loosely into a jar, covered with an extracting
liquid like alcohol and water, glycerine, vinegar, or olive oil.
2. After absorbing herbal properties for a time, use a press to separate the liquid from

3. Finally pressing the herbs by hand using a tincture press and the exhaust left is further
expressed using a muslin cloth to extract and remaining extract.

WORKING
1. Set up the electrode stand and fit the pH electrode
in it.
2. Carefully remove the protective rubber cap from the filling hole of electrode.
CALIBRATION OF ELECTRODE
1. The electrode should be calibrated before beginning measurements or when pH
readings are doubtful. Once the calibration has been carried out, do not change the
electrode without fresh calibration.
2. Connect the combination pH electrode to the input socket, wash it with distilled water
& switch ON the instrument.
3. Dip the electrode in 7pH Buffer solution.
4. Set the “TEMPERATURE” “C” control to the buffer solution temperature.
5. Set the Function Selector Switch to ‘pH’ position and adjust with ‘CALIBRATE’
control till the digital display shows the precise pH value of the buffer solution.
6. Now move the Function Selector Switch to “STAND BY”.
7. Remove the electrode from the buffer solution & wash it with distilled or de-ionised
water.
8. Dip the combined electrode into another buffer solution (say 4 pH)
9. Set the ‘TEMPERATURE’ “C” control to the temperature of the selected buffer
solution.
10. Set the Function Selector Switch to pH position. Adjust the “Slope” correction
Control, at the front panel until the display shows the pH value of the selected buffer
solution. Check that the correct readings are obtained with both the buffer solutions
without further adjustment.

Tap/cock - This helps us to get the water from the source and

WORKING
1. Take a bottle whom you wish to wash.
2. Get some detergent /chemical in the machine.
3. Switch ON the machine and regulate the speed as per requirement (generally machine
is pre settled on the desired speed)
4. Attach the tap from below to the water source and connect some hose/pipe with it to
get the water.
5. Now get some water in the bottle you wished to wash and then put the bottle near the
brush spindle.
6. Your washing is done.
PRECAUTIONS
1. Take care of the electrical hazards.
2. Take care of your hands during washing at it can be hurt by the spindle.
3. Kindly keep the machine clean “No Use” condition.

WORKING
1. Before use ensures that the pans are clean, stirrups on the knife edges and pointer on
the zero mark of the scale of spring balance.
2. Powdered chemicals and liquids should be weighed on a watch glass or in a weighing
tube, but not directly on the pan.
3. See that the beam is not in motion while putting in or removing an object on a weight
from the pan.
4. Always use forceps in lifting the weights.
5. Put weights on the right hand pan and the object on the left hand pan.
6. Hot containers should not be weighed.
7. Start weighing with a weight heavier than the object.
8. The last reading should be taken after closing the balance.
9. Close the spring balance after you have finished.
PRECAUTIONS
1. Place the balance in proper and fixed position.
2. Before starting the weighing, check the movement of pointer are equal to both the
side.
3. Check the pans, stirrups are in proper position on beam.
