Topic:Investigate Factors Affecting Diffusion

Activity#1: Investigate Factors Affecting Rate of Diffusion

Experiment: Size Matters: Uncovering the Dynamics of Diffusion with Agar Cubes

Objective:
To investigate how the surface area of cells affects the rate of diffusion of a substance through
them.

Materials Required:

● Agar jelly prepared with a small amount of ammonium hydroxide and colored with
cresol red
● Sharp knife
● Plastic or blunt-ended forceps (tweezers)
● Ruler (measuring in mm)
● Two large test tubes or similar glass tubes (with a diameter greater than 1 cm)
● Stopwatch
● Dilute hydrochloric acid (handle with care and wear safety glasses)
● Cold water (for spill clean-up)
● Graph Paper

1. Prepare Agar Cubes:


○ Cut one cube of agar jelly with sides of 1 cm into eight smaller cubes, each with
sides of 0.5 cm. This will give you a large cube and eight smaller cubes.
2. Set Up the Test Tubes:
○ Place the large 1 cm cube into one test tube.
○ Place the eight smaller 0.5 cm cubes into the other test tube.
3. Add Acid:
○ Pour equal volumes of dilute hydrochloric acid into both test tubes, ensuring that
all the cubes are fully submerged.
4. Start the Experiment:
○ Start the stopwatch as soon as the acid is added to both test tubes.
5. Observe and Record:
○ Monitor the color change of the agar cubes as the acid diffuses into them.
○ Stop the stopwatch when the cubes in each test tube turn fully yellow.
○ Record the time taken for the color change to occur in each test tube.
○ Fill in the recorded time for each cube size in the table below.

Plot a Graph: Using the data from the table, plot a graph to show the relationship between
cube size and the time taken for diffusion. On the x-axis, plot the cube size, and on the y-axis,
plot the time taken for diffusion.

Analyze: Analyze your graph to understand how the size of the cube affects the rate of diffusion.

1. The jelly cubes contained a weak alkali and cresol red, which is an indicator. Explain why

Note: In the previous experiment, you used cubes of alkaline agar jelly and an indicator , so that
you could measure the time taken for an acid to diffuse through the cubes.
Now you are going to plan how you can use this technique to investigate other factors.
You may be able to carry out the experiment that you plan.

Instructions:
Apart from surface area, diffusion is also affected by:
a) Temperature
b) Concentration gradient
c) Distance
Choose one of these variables to investigate

2. The variable that you change in your experiment is called the independent variable. What is
the independent variable in your experiment?

3. Think about how you can change your independent variable. Decide how many different
values you will have.

4. The variable that you measure, as you collect your results, is the dependent variable. What is
the dependent variable in your experiment?

5. Describe how you will measure your dependent variable.

6. It is important that no variables, other than the one you are investigating, affect your
experiment. Which variables will you standardise (keep the same) in your experiment? How will
you do this?

7. Write a short, clear description of how you will do your experiment.

8. Draw a results chart that you could use to fill in your results. Include headings, but do not put
in any results (unless you are able to actually do your experiment, of course).

9. Create a particle model diagram to illustrate the expected outcome of your experiment.
