TECHNICAL PROCEDURES IN PERFORMING LABORATORY TESTS: HEMATOLOGY
1) Complete Blood Count
Specimen: Whole Blood
Check if the information in the test tube label matches with that of the requisition form. Invert the tube 8-10 times. Ensure the analyzer is turned on. Remove the tube stopper, insert the sample tube in the sample probe. Press the button behind the probe, and allow the probe to aspirate sample. Remove the sample tube as soon as you hear a beep sound, this indicates that enough sample has been aspirated. Wait until the results are flashed on the screen. Print out results.
2) Clotting time (Slide method)
Sample: Fresh capillary blood
Collect a small drop of capillary blood in the capillary tube. Gently expel blood onto the glass slide, forming a small drop. Tilt the slide back and forth, then rotate it gently to spread the blood evenly. Observe the blood drop for clot every 30 seconds. Record the time it took for the blood to form fibrin clots.
3) Bleeding time
Sample: Fresh capillary blood
Ensure the patient has not taken medications affecting bleeding time. Make a small prick on the selected site using the sterile lancet. Allow blood drop to form on the site. Wipe the blood drop on the site every 30 seconds, until the bleeding stops. Record the time it took for the bleeding to stop.
4) ABO and Rh Typing (Forward Typing)
Mark a clean glass-slide as anti-A, anti-B, and anti-D.
Place 1 drop of blood on each marked space. Add 1 drop of anti-A serum to the blood on the space marked with anti-A. Add 1 drop of anti-B serum to the blood on the space marked with anti-B. Add 1 drop of anti-D serum to the blood on the space marked with anti-D. Using an applicator stick, mix each side of the reaction. Rock the slide back and forth and observed for agglutination. Record the observation. Interpretation of results: o Agglutination with anti-A: Blood Group A o Agglutination with anti-B: Blood Group B o Agglutination with both anti-A and anti-B: Blood Group AB o No agglutination with both anti-A and anti-B: Blood Group O o Agglutination with anti-D: Rh positive o No agglutination with anti-D: Rh negative
