Investigating the effect of PH on Amylase

Iodine
·
solution is dropped in rows on the tile .

·
The test tube is labeled with the pH tested.

·
2 cm3 of amylase is placed using the syringe in the tube .
·
Another tube is used to add 2 cm3 of starch solution to the

amylase and buffer solution , the stopwatch is started whilst

the solution is mixed pipette.

using a

·
The mixture is dropped on the first drop of iodine after 10s
which should turn blue-black.

·
Another drop of mixture is placed on the second drop of iodine
after wait another 10s .

. This process is repeated every los until the solution remains

orange-brown .

· The experiment is repeated at different pH values

The less time the iodine solution takes to remain orange-

- brown , the quicker all the starch has been digested and so the

Enzyme action better the enzyme works at that pH.

·
lock and key hypothesis .

how enzymes work :

1 .

Enzymes and substrates randomly move in the solution .

.
2 When an enzyme and its complementary substrate randomly collide , an enzyme
-
substrate

complex forms and the reaction occurs.

.
3 A product forms from the substrate then released from the active site. The enzyme is

unchanged and will go on to catalyse further reactions .
