Primarily I would thank God for being able to complete this


project with success. Then I would like to thank my biology
teacher Mrs. Smriti Fuley whose valuable guidance has been
the ones that helped me patch this project and make it full
proof success her suggestions and her instructions has served
as the major contributor towards the completion of the
project.
Then I would like to thank my parents and friends who have
helped me with their valuable suggestions and guidance has
been helpful in various phases of the completion of the
project.
Last but not the least I would like to thank my classmates
who have helped me a lot.

INTRODUCTION

All living organisms have DNA, which is short for deoxyribonucleic acid; it is
basically the blueprint for everything that happens inside an organism’s cells.
Overall, DNA tells an organism how to develop and function, and is so
important that this complex compound is found in virtually every one of its
cells.
Each of your cells will have DNA inside of it (with some rare exceptions, such as
mature red blood cells in humans). Each cell has an entire copy of the same set
of instructions, and this set is called the genome. Scientists study DNA for
many reasons: They can figure out how the instructions stored in DNA help
your body to function properly. They can use DNA to make new medicines or
genetically modify crops to be resistant to insects.
DNA extraction from strawberries is a well-known procedure that allows
students to visualize DNA and to recognize that living things contain DNA.
Many similar protocols exist, and most involve simple materials that can be
readily obtained
We will use an extraction buffer containing salt, to break up protein chains
that bind around the nucleic acids, and dish detergent which helps to dissolve
the phospholipid bilayers of the cell membrane and organelles. This extraction
buffer will help provide us access to the DNA inside the cells. DNA is not
soluble in alcohol. The colder the alcohol, the less soluble the DNA will be in it.
Thus make sure to keep the alcohol in the freezer or on ice.

AIM: TO EXTRACT DNA FROM STRWABERRIES
MATERIALS:

1. 2 Strawberries (fresh or frozen).


2. Re-sealable Plastic Bag.
3. 2 tsp Dish Detergent.
4. 1 tsp Salt.
5. ½ Cup Water.
6. 1 Coffee Filter.
7. 2 Plastic Cups.
8. ½ Cup Cold Rubbing Alcohol.
9. Coffee Stirrer.

EXPERIMENTAL PROCEDURE

1. Pull off any green leaves on the strawberry.


2. Put the strawberries into the plastic bag, seal it and gently smash the
strawberries for about two minutes. This starts to break open the cells
and release the DNA.
3. Mix together 2 teaspoons of detergent, 1 teaspoon of salt and 1/2 cup of
water in a plastic cup. This will be your DNA extraction liquid.
4. Pour the DNA extraction liquid into the bag with the strawberries (make
sure the bag is big enough; quart-sized works well!). This will further
break open the cells.
5. Reseal the bag and gently smash for another minute. Avoid making too
many soap bubbles.
6. Place the coffee filter inside the other plastic cup.
7. Open the bag and pour the strawberry liquid into the filter. Twist the
filter just above the liquid and gently squeeze the remaining liquid into
the cup.
8. Pour an equal amount of cold rubbing alcohol as there is strawberry
liquid down the side of the cup. Do not mix or stir.
9. Within a few seconds, watch for a white cloudy substance (DNA) to
develop in the top layer. You have just isolated the DNA from the rest of
the material contained in the cells of the strawberry.
10.Tilt the cup and pick up the DNA using a coffee stirrer.

SAFETY PRECAUTION

Avoid eye contact with this material. Wear chemical safety


goggles. Use chemical gloves as needed. Use clean protective
body covering clothing as needed to minimize contact with
clothing and skin.

CONCLUSION

DNA was successfully extracted from a strawberry,


Demonstrating the process a real life scientists would
possibly extract DNA from cells. The lab was intact successful
for the group and I were able to extract a visible amount of
DNA from the mixture.
