What is the use and composition of a running buffer? Sample buffer? (4 points)

Why is a semi log graph always crucial in accurately determining DNA size during agarose gel
electrophoresis? (6 points)

What are the two techniques that are widely used to perform DNA fingerprinting analysis?
Briefly discuss the concept behind those techniques stating the advantages and disadvantages
of each. (6 points)

You are studying a rare autosomal recessive human disease that is fully penetrant in adults. You
have identified a restriction fragment length polymorphism (RFLP) that is 10 map units from
the disease gene. Consider the following pedigree and Southern blot showing inheritance of the
RFLP. Individual D is too young to show the disease trait.
Using the above pedigree and ignoring the RFLP data, what is the probability that individual
D will be affected by the disease as an adult? (3 points)

Considering all the data, give the genotypes for the individuals F, C, and E. Define any
symbols used (4 points)

A common epistatic trait is albinism (lack of pigment). You are all familiar with white
laboratory rats. These are albino rats. Rats may also have agouti (grey) or black coats. These
are determined by the alleles G and g. The genes for albinism are a separate set of alleles.
These genes are either A or a.
Key: G = gray g = black A = color or pigment is present a = color or pigment is absent

What color would a rat who is GGaa be? How about if the rat was genotype ggAa? (2 points)

Try writing the genotypes for the following rats. (2 points)

• Two black rats have albino and black offspring. Please show all the genotypes of the

pigmentation. Please show all the phenotypes of the progeny of this cross (with the
