35. Draw a neat labelled diagram of sectional view of the mammary gland.

36. Schematically represent the inheritance of flower colour in snapdragon and draw conclusions.
It is an inheritance in which heterozygous offspring shows intermediate character b/w two parental
characteristics. E.g. Flower colour in snapdragon (dog flower or Antirrhinum sp.) and Mirabilis jalapa
(4’O clock plant).
Here, cross between homozygous red & white produces pink flowered plant. Thus phenotypic &
genotypic ratios are same.

Phenotypic ratio= 1 Red: 2 Pink: 1 White


Genotypic ratio= 1 (RR): 2 (Rr): 1(rr) This means that R was not completely dominant over r.

38. Describe the steps involved in DNA fingerprinting technique.


1. Isolation of DNA (from any cells or blood stains, semen stains, saliva, hair roots, bone, skin etc.).
2. Digestion of DNA by restriction endonucleases: long strands of DNA are cut with specific
molecular scissors (REN) into desired lengths.
3. Separation of DNA fragments by gel electrophoresis: (The DNA fragments are separated
according to their length and arranged on using Agarose gel electrophoretic technique.)
4. Transferring (blotting) DNA fragments to synthetic membranes such as nitrocellulose or
nylon: (Southern blotting technique is used to blot the fragments of DNA from gel slab to
nitrocellulose paper.)
5. Hybridization using radioactive labelled VNTR probe. (The single stranded DNA are made to
form complementary strands with known radioactive VNTR probes. The other strands which do not
hybrid are washed with water.)
6. Detection of hybridized DNA by autoradiography: (When nitrocellulose paper is subjected to X-ray,
DNA bands with radioactive labeled probes appear as dark bands. Such bands are unique to a person and
vary from person to person like the finger prints.)

a) What is EcoRI? (1M)


EcoRI is a Restriction Endonuclease Enzymes (‘Molecular Scissors’). EcoRI comes from

b) How is the action of exonuclease different from that of endonuclease? (2M)
They break or cut the 5' or 3' ends of DNA molecule i.e. they remove nucleotides from terminal
ends of DNA in one strand of double helix.
c) How are ‘sticky ends’ formed on a DNA strand? Why are they so called? (2M)
Restriction enzymes cut the strand a little away from the center of the palindrome sites, but
between the same two bases on the opposite strands. This leaves single stranded overhanging
stretches at the ends. They are called sticky ends.
