Experiment No.

1 DNA ISOLATION Date:

Aim – To isolate DNA from spinach leaves.

Theory – DNA is the nucleic acid which contains genetic instructions that
play important role in development and functioning of living organisms.

Requirements – 50 gm spinach leaves, 200 ml cold water, quarter tsp salt,


meat tender riser – papaya paste / pineapple juice / contact lens solution,
90% ethyl alcohol, blender, timer, strainer, 2 tbsp detergent powder,
measuring glass, wooden stick.

Procedure – i. 50 gm of spinach, quarter tsp of salt and 200 ml of


cold water is taken in a blender and blended for 20 sec, after
which it is strained into a measuring glass container.
ii. 2 tbsp of detergent powder was added and mixed gently
allowing it to sit for 10 minutes after which it is poured in
few test tubes about one-third full.
iii. to each of the test tubes, pinch of meat tenderiser was
added and stirred gently.
iv. equal amount of chilled alcohol was added and allowed
to wait for 3 minutes with alcohol forming a layer on top.
v. clump of white stringy mass seen at the junction of liquid
mixture.
vi. with the help of wooden stick DNA is spooled out of the
alcohol.

Observation – clumps of white, thin fibrous DNA is observed.

Inference – DNA is the blue print for life and can be isolated through
physical and chemical methods.

Precautions – i. enough time should be given for each step.


ii. mixing or stirring should be done gently.
