Motility Test
PURPOSE
Determines whether enteric organisms are motile.
PRINCIPLE
Bacterial motility is evident by a diffuse zone of growth in
semisolid agar extending out from the line of inoculation. Some
organisms grow throughout the entire medium, whereas others
show small areas of growth out from the line of inoculation.
Some tests include triphenyltetrazolium chloride (TTC). TTC in
its oxidized form is colorless. As bacteria grow in the presence
of TTC, it is absorbed and reduced to the insoluble red-colored
pigment formazan.
If TTC is present, growth is indicated by the presence of the red
color, and as motility occurs, small to very large regions of color
can be observed around the area of inoculation. If no TTC is
present, growth is indicated by cloudiness.
