The study guide outlines key techniques and concepts related to the secretory pathway, including pulse-chase experiments, the role of temperature-sensitive proteins, and the function of various vesicles. It covers both early and later stages of the secretory pathway, detailing mechanisms of protein sorting, retrieval, and endocytosis. Additionally, it explores the differences between endocytic and exocytic vesicles and the pathways of degradation.

Study guide/guiding questions for Chapter 14:
Techniques for studying the secretory pathway.
1) Design and interpret a pulse-chase experiment to follow a secretory protein 2) Describe the temperature sensitive VSV Glycoprotein and its importance in the protein trafficking field. 3) How is the compartment specific modification of carbohydrate moiety of glycoproteins and its useful to study protein secretion? 4) What are endoglycosidases? What are Lectins 5) Design and interpret experiments utilizing Endo H and/or Endo D to look at the progress of a particular glycoprotein along the secretory pathway. 6) Why were yeast genetic studies so useful to identify and confirm the protein components that participate in the secretory pathway in humans? 7) How would you test the cytosolic components needed for the process of vesicle budding, transport and fusion?
Early Stages of the secretory pathway:
1) Describe the components and functions of COPII, COPI and clathrin transport vesicles. 2) Describe the mechanism of budding, coat shedding and fusion to target membrane. 3) What (and which proteins) controls the coat polymerization and de-polymerization. 4) Describe how lumenal and membrane cargo proteins are sorted into specific vesicles. 5) Describe the consequences of inactivating mutations in Sar1, v-SNARE, Rab, etc. 6) How are ER-resident lumenal and membrane proteins retrieved or kept in the ER? 7) How are Golgi-resident proteins retrieved from trans- to medial- to cis-Golgi?
Later stages of the secretary pathway:
1) What is the main branching point in the secretory pathway? 2) What protein is required for the pinching-off process from clathrin coated vesicles? 3) Describe the complete process of targeting enzymes to the lysosomes 4) What mechanism insures the rapid release of some hormones or enzymes from secretory cells? 5) How are proteins destined to the apical aspect of the cell sorted? 6) What is transcytosis?
Receptor-mediated endocytosis and pathways of degradation:
1) How do endocytic vesicles differ from exocytic vesicles coming from the trans-Golgi network? 2) What are the sorting signals present in plasma membrane receptors that lead to receptor-mediated endocytosis? 3) What is the final destination of the endocytic pathway for the majority of cargos? 4) What are endosomes? Describe the autophagy pathway?
