Aim:

prepare a temporary mount to observe pollen germination


Material required:
flowers of various type i.e, Hibescum, Plide plane, cavity Salide, mong brush. needles
coveselip, Broker, dropper, micmscope & nutrient medium.
Preparation of Nutrient medium:
Sucrose: 10 gm
Boric Acid: 10 gm
KNO3: 10gm
MgSO4: 10gm
dissolve in 100 ml of water.
THEORY:
Pollen grain represents the male gamete and have an amazing architecture size shape,
colors and design. It has a 2 layered oils wall, outer exine and inner intine. The area
where sporopollenin is absent in germ pure.
Procedure:
Take a few drops of above nutrient solution in a clean cavity slide Now take some
flowers of locally available species with a help of a brush, dust a few pollen grain from
the flowers into the cavity containing culture solution cover it with covership place the
slide undisturbed for sometime, Observe the slide after every 15 minutes to see the
Pollen grain germination and growth of pollen tube. Repeat the experiment with pollen
of other flowers.
Observation:
In nutrient medium, the pollen grains germinates The tube cell enlarges and come out of
the Pollen grain through the germ pores to form Pollen-tube The tube nucleus descends
to the tip of the pollen tube Generative nucleus also through it and then divides into two
male gamete.If pollen grain germinate then the pollen is viable and if they do not
germinate then they are not viable.
Conclusion:
The pollen germinate in nutrient medium includes the growth of pollen tube. In some
species of flower, the percentage of pollen germinates in layered in Somecase, the
pollen do not germinate at all.
Precaution: use clean and dry slide to observe pollen grain flower should be freshly
plucked. The pollen from one species should not mix with other pollen
