Topic: Cells: The Basic Units of Life (Molecular make-up)

AIM OF THE INVESTIGATION: To determine the effect of temperature on the activity of catalase
(found in raw/uncooked potato).

1. Formulate a hypothesis for the investigation. (2)

A high / increase in temperature increases/ decreases the activity of catalase 


Or
A low temperature will increase/decrease the activity of catalase

An increase in temperature has no effect on the activity of catalase

2 Identify the following variables:

2.1 Dependent: activity of catalase  (1)

2.2 Independent: temperature  (1)

3. Identify ONE controlled variable:

 size of the potato/amount of potato paste(1ml), 


 amount/volume of Hydrogen peroxide (5ml), 
 amount / volume of water and crushed ice in each beaker ( 150ml)
 Same beaker size
 Same incubation period/ observation time

4 Tabulate the results obtained during the investigation.

6.2 What conclusion can be drawn from QUESTION 6.1?

Low temperature of 20C slows down catalase activity, whilst 280C is the optimum (1)
temperature for the highest catalase activity and catalase activity decreases at
temperatures of 900C

No , the denatured enzyme active site cannot go back to its original shape for the
substrate to fit

8. Consider the scenarios below. Mention AND explain how the results in the test tubes
would have differed if:

8.1 Sodium hydroxide was added to the hydrogen peroxide. (2)

It would increase the pH and denature the enzyme/catalse causing the activity of
the catalase to slow down 

8.2 The potato added wasn’t grinded into a paste. (2)

The surface area exposed to hydrogen peroxide would be smaller , less catalase
Activity/  The foam would have formed at a slower rate .
