ABO Rh PROCEDURE
A. SPECIMEN
- Anti-coagulated Whole blood (Lavender Top)
- Serum
B. REAGENTS
- Anti-sera A, Anti-sera B, Anti-sera D
- Red Cell Suspension – A1 and B red cell. (Commercially prepared or laboratory
prepared from a 3% to 5% red cells suspension of pooled from known blood type “A”
and known blood type “B” donor red cell)
C. SUPPLIES
- Test tubes
- Calibrated plastic pipette (0.5 – 5 uL)
- Buffered Normal Saline Solution (NSS)
- Serologic Centrifuge
D. PROCEDURE
1. Label five (5) clean test tubes respectively as: Anti-A, Anti-B and Anti-D for forward grouping
and Known “A” and Known “B” (A1 cells & B cells) for reverse grouping.
2. Place 1 drop each of Anti-A, Anti-B and Anti-D in their respective labeled clean test tube.
3. Place 1 drop (approx.. 50 uL) each of Known “A” (A 1 cells) and Known “B” (B cells) in their
respective labeled clean test tube.
4. Add 1 drop of patient’s red cell suspension (3% - 5%) to test tube labeled Anti-A, Anti-B and
Anti-D.
5. Add 2 drops of patient’s serum / plasma to test tubes labeled Known ”A” and Known “B”.
6. Mix gently the content of all tubes and centrifuge them for the calibrated spin time.
7. Gently re-suspend cell buttons and examine them for agglutination.
8. Read, interpret, and record the test results accordingly.
