Practical 4 – Observation of GUS expression in transgenic rice

1. Practical objectives:
The purpose of this practical is to observe the GUS expression in transgenic rice in order
to characterize the localization of the given gene.
2. Biological Material:
- Transgenic Rice containing GUS gene fused with a specific promoter (of gene A) was
stained by GUS staining solution (X-Gluc (5-bromo-4-chloro-3-indolyl-glucuronide) – the
substance of β-glucuronidase and other components as appropriate buffer of β-
glucuronidase)
- WT rice was stained by the same GUS staining solution
3. Equipment
- Microtome machine to section the rice roots;
- Glass slides;
- Microscopy.
4. Procedure
- Observe the transgenic rice (compared to the WT rice) to determine the organ in which
GUS is expressed;
- Section transversely the region where GUS is expressed;
- Observe the cross section under microscopy to determine the tissues where GUS is
expressed;
- Give the conclusion about function of gene A in rice.
