Fluorouracil, known commercially as Drucil, is a pyrimidine analog and a
fluorinated derivative of uracil.

Uracil Fluorouracil(5-FU)
5-fluoro-2,4(1H,3H)-pyrimidinedione

Composition: Brand Names:


250 mg of Fluorouracil in 5 ml
of solution Drucil, Adrucil,
500 mg of Fluorouracil in 10 Efudex, Carax, Tolak
ml of solution
Injectable solution
Mechanism of action
Pharmacokinetics:
5-fluorouracil is rapidly metabolized in the liver to produce biologically inactive metabolite which are eventually converted to
carbon dioxide, eliminated by the lungs. 80 % of can be accounted for by conversion to respiratory CO2 within 12 hr of
administration. 15 % of the drug is excreted through urine.

Therapeutic indications
Fluorouracil is indicated alone or in combination for Carcinoma of the -

Colon Liver Breast Lung Bladder Cervical


or Carcinoma
rectum
Dosage & Administration
Usual Dose:
• Initial Course: 12 mg/kg once daily for 4 days

• Maintenance Therapy: 5–10 mg/kg weekly

• Cervical Cancer (with Cisplatin): 1 g/m² IV on day 1, repeated every 21 days

• Pediatric Use: Safety and efficacy not established


Combinations in Treatment:
Administration • With radiation and other anticancer drugs
