APSI Poster (HPTLC)

This document summarizes a study analyzing the phytoconstituents in extracts of Litsea polyantha Juss bark using thin layer chromatography (TLC) and high performance thin layer chromatography (HPTLC). Preliminary tests found sterols, phenols, flavonoids, terpenes and alkaloids in the extracts. TLC and HPTLC were used to separate and analyze the components in petroleum ether, chloroform, ethyl acetate and methanol extracts. Multiple spots were detected in each extract. Absorption maxima were identified for different spots. The analyses can help determine active compounds in the bark which is used traditionally for conditions like diarrhea.

OBJECTIVES FOR UNDERTAKING THE WORK Absorption Maxima ( λ max

) of different Spots observed on HPTLC
The bark of Litsea polyantha Juss (Lauraceae) has a long history of Extract Petroleum Chloroform Ethyl acetate Methanol
medicinal use among the traditional healers of Oraon and Munda ether
community of Jharkhand. In Folklore medicines – The bark of L.
polyantha Juss is reported to be used for diarrhea. Powdered bark and Spot Hot Hot Hot Hot
roots are used in external applications for pains, bruises and 1 306 271 219
contusions; they are also used for fractures in animals. The present
study is carried out to analyze the Phytoconstituents present in Litsea 2 285 317 282
polyantha Juss. Bark extracts by TLC & HPTLC.

EXTRACTION 4 257 220

The dried and powdered plant materials were subjected to successive 5 372
hot extraction with Petroleum Ether, Chloroform, Ethyl Acetate and
Methanol to afford respective extracts. 6 255

PHYTOCHEMISTRY 8 206
A preliminary phytochemical investigation of the extracts revealed the
presence of Sterols, Phenols, Flavanoids, Terpenes and Alkaloids. 9 233 301 200

MATERIALS AND METHODS 11 209
Instrument: . Camag HPTLC
Band Applicator: Linomat IV automated spray-on band applicator
equipped with a 100-GL syringe
HPTLC Plates: Precoated Silica Gel 60 F254 Plates (20 x 10 cm)
Scanner: Camag HPTLC Scanner 3
Software: CATS 4.0

PROCEDURE
Petroleum Ether (PELP), Chloroform (CELP) , Ethyl acetate (EELP)
and Methanol (MELP) Extracts were analyzed by TLC & HPTLC

The spots / bands were monitored on UV light (254 nm & 365 nm),
iodine chamber and by using various spraying reagents for detection of
the phytoconstituents..
