I cannot believe my life. Every day, I wake up, with not much sleep, knowing of the

amount of chloroplast present at an absorbance reading of 600 nm? To answer this
question, we used centrifugation to separate out Pellet 1, Pellet 2, and Supernatant 2,
and then we tested each of them at 600 nm three times to determine which one
contained the most chloroplast. Based on our results for wet lab 1 we hypothesized
that Pellet 2 would contain the most chloroplast. We predicted this because in wet lab
1, we learned that a homogenized solution contains all organelles, Pellet 1 contains
large amyloplasts, and Pellet 2 contains chloroplasts and small amyloplasts

Abstract
The purpose of the experiment was to isolate plasmid DNA, followed by restriction
digestion using restriction endonucleases and then visualizing the digested fragments
after subjecting to gel electrophoresis. Plasmid DNA (pSP72 DNA) was isolated
from Escherichia coli KAM32 (E.coli) cultures using the QIA prep miniprep kit and
then subjected to restriction digestion by EcoRI and HindIII. The restriction digested
DNA was then loaded into the wells of 0.7% agarose gel and subjected to
electrophoresis. It can be concluded from our results that our plasmid DNA isolation
was successful and the restriction digestion results were partially in agreement with
our hypothesis.

containing their own origin of replication. Hence, they are capable of replication
independent of the chromosomal DNA in bacteria. Plasmids present in one or
more copies per cell, can carry extra chromosomal DNA from one cell to another
cell and serve as tools to clone and manipulate genes. Plasmids used exclusively for
this purpose are known as vectors. The genes of interest can be inserted into these
vector plasmids creating a recombinant plasmid. Recombinant plasmids can play a

Senior Management Retreat
I was tasked with developing an agenda topics for the Senior Management retreat to
be held on May 31, 2016. In order to identify the topics for discussion I needed to
identify what was the current state of the company s profitability and what the
company is doing to be a socially responsible organization.
First, I completed a thorough review of the company s current business practices and
conducted several focus groups with employees to discuss their top five ideas and
suggestions for improving their productivity, commitment to the company and overall

When working, how much do you value what you do? For me, a part time worker
in college just trying to have some extra cash on hand, I don t seem to value my
job that much. Now as I pursue my career and obtain my end goal, I will value my
job as much as I valued the education it took to get there. My mind set seems to be
